Resumo:

Pancreatic ductal adenocarcinoma (PDAC) is the most aggressive pancreatic cancer, representing 85% of cases. Key mutations, such as in KRAS (90% of patients) and TP53, drive its progression, alongside epigenetic changes like DNA methylation and histone modifications. The tumor microenvironment (TME) plays a major role in therapy resistance by shielding cancer cells. This study tested a novel therapy combining epigenetic inhibitors (e.g., Entinostat), chemotherapy (gemcitabine), and siRNA targeting YAP-1 and FOSL1. This approach disrupts stroma-tumor interactions, reduces tumor growth, and enhances immune responses. Using a 3D organoid model, the therapy showed promise in overcoming PDAC resistance and improving treatment efficacy.
