ABSTRACT:
Polyenic macrolactams (PMLs) are a family of natural products with more than 50 members reported and interesting biological activities. Their structures have complex motifs like conjugated polyenic chains, medium size macrocycles (20 – 34 atoms) or aminosugars linked to asymmetric carbons. This thesis describes the total synthesis of four PMLs. The structure of muanlactam proposed by computations has been synthesized and the structure described upon isolation was corrected. Three isomers of the macrolactam JBIR-150 were synthesized but their spectroscopic data did not match those of the natural product. This work serves the purpose to understand and try to elucidate the structure of the natural product. Additionally, the octahydronaphthalene core of sagamilactam was synthesized via a bis-TADA through a completely stereoselective Si-exo approach.
